All off-target sites produced using HiFi Cas9 and a 20-nucleotide guide RNA were detected by all the other methods of identification, excluding the SITE-seq method. A characteristic of the majority of OT nomination tools was high sensitivity, with COSMID, DISCOVER-Seq, and GUIDE-Seq showing the best positive predictive values.
